Strikes from James Kaye and Alon Hershkorn saw Brixton cause one of the biggest shocks in MSFL history as they dumped former winners Neasden out of the cup.
The tie was goalless for 63 minutes when Kent raced clear on goal only to be hauled down by Matt Sofier, who was sent off.
Up stepped substitute James Kaye to take the free kick just outside the area with a looping shot which gave their keeper no chance.
Hershkorn settled it in the 75 minute - scoring with his head from a corner.
Delighted Brixton boss John Nitka told JC Sport: "What a result a match. It could have gone either way in the first half."
"It was a fantastic performance by us and are looking forward for our second encounter with Neasden next week.
"Even with 10 men Neasden pressed but our defence held them out."
Neasden joint-manager Darren Yarlett told JC Sport: "We lost in a game that should have been over by half-time.
"Once again, we created chance after chance and failed to convert. Our forwards need to take a greater responsibility. We were hurt today by players away or injured but that's no excuse.
"Credit to John Nitka and his lads who worked hard all day, and a mention to Warwick Blake today who showed why he is by far the best referee in the league.
"Football can be a cruel and kind game. We lost today and my players will be hurting but they can put it right next week when we play them again in the Peter Morrison Trophy."
