The Thornes made it seven out of seven in a thriller against title rivals UJIA at Chase Lodge.
Joint-player-manager Craig Sandford led the way with a double. Daniel Lassman and a penalty from James Rubin sealed the win.
UJIA's goals came through Gavin Rhodes and Toni Plaskow.
New signing Fabio Revieccio did not play for the Thornes.
Glenthorne joint-player-manager Craig Sandford told JC Sport: "We showed great quality today and even when our heads could have dropped, we kept positive and believed in our ability.
"At 1-0 down we made sure we got level quickly and when UJIA scored to make it 2-2 we knew we had to battle to get the three points.
"A real professional performance against a very well organised UJIA team. Credit to them for going all the way to the final whistle and trying to get something out the game."
UJIA player-manager Darren Rolfe told JC Sport: "We gifted them two goals with sloppiness which isn't acceptable.
"It was a fair result in the end, but I'm disappointed we didn't take anything out of this game as there was a real opportunity today. Glenthorne are a good side but they're beatable."
